El protagonista de esta serie es Adrian Monk, un ex-detective del departamento de homicidios que en la actualidad trabaja como asesor para la policia de San Francisco. Este tuvo que abandonar el cuerpo ya que no estaba preparado mentalmente para poder trabajar, su mujer había muerto en un coche bomba. Él era un ser peculiar ya de por sí, pero la muerte de su esposa, desencadeno en él un sinfin de miedos. Trudy, su mujer, era lo unico que le hacia ser normal y desde el momento en que ella falleció, se puso en búsqueda de los culpables. Esta considerado como el mejor detective del mundo pese a lo estrafalario que pueda parecer tanto él como sus metodos para pensar. Debido a su problema psicologico, va acompañado siempre de su asistenta (Sharona en las 3 primeras temporadas y Natalie en las restantes) la cual tendrá que cuidar de su jefe como si de un niño pequeño se tratara, y soportando sus manias y fobias(algunas de ellas absurdas, como su temor a la leche). Juntos trabajan como asesores para la policia, que esta comandada por el capitan Stottlemeyer y su teniente Randy Disher. Entre los 4 personajes principales resolveran en cada episodio los crimenes que se planteen, mientras sigue una línea argumental contínua que es la de Monk investigando el caso de su mujer, el único crimen que no ha podido resolver.
